Composition & Mechanism of Action

• Active Ingredient: Atovaquone USP 250 mg per tablet.

• Pharmacological Action: Atovaquone acts as a competitive structural analog of ubiquinone (coenzyme Q), blocking the electron flow through the cytochrome bc₁ complex in protozoal mitochondria. This collapses the parasite's mitochondrial membrane potential, inhibits nucleic acid synthesis, and triggers rapid protozoal death.

• Clinical Dosing Regimen (General Veterinary Literature Guidelines):

  • Dogs (Babesia gibsoni): Typically 13.5 mg/kg body weight orally every 8 hours (three times daily), or 10–15 mg/kg as determined by the veterinary physician, administered for a continuous 10-day cycle alongside Azithromycin (10 mg/kg once daily).

  • Cats (Cytauxzoon felis): Typically 15 mg/kg body weight orally every 8 hours (three times daily) alongside Azithromycin (10 mg/kg once daily) for 10 days.

• CRITICAL Bioavailability Rule:

  • Administer with a Fatty Meal: Atovaquone is highly lipophilic. Administering the tablet alongside a small, fat-rich meal (e.g., a spoonful of canned meat, egg, or cottage cheese) increases oral absorption up to threefold. Giving on an empty stomach drastically lowers blood levels and reduces clinical efficacy.

• Follow-Up Testing: Post-treatment PCR testing is strongly recommended 30 to 60 days following completion of therapy to confirm negative parasitemia status.
